Il commissario Giovanna Scalise, trasferita a Roma per motivi di sicurezza, dopo le dimissioni da capo del distretto del commissario Grimaldi (personaggio mai apparso nella serie, ma menzionato dal sovrintendente Antonio Parmesan), cerca di arrestare i mandanti e i killer del marito Ferdinando, giornalista ucciso dalla mafia siciliana. Nella sua ricerca dei colpevoli si troverà a rischiare più volte la vita, insieme a quella dei suoi figli.
Il commissario, che assume subito le particolarità del capo autoritario ed esigente, inizia a conoscere questa sua nuova squadra con la quale non si aprirà molto nella prima parte della serie. L'ispettore del distretto, Roberto Ardenzi scoprirà il passato della Scalise, mettendo al corrente l'intera squadra. La squadra, determinata subito nel proteggere il suo commissario, si accorgerà di aver aperto un conto difficile da chiudere. A farne le spese sarà Nina Moretti, impegnata durante la scorta alla figlia della Scalise viene uccisa e vengono coinvolti Manrico e la figlia del commissario. Tale morte porterà sgomento negli agenti del X Tuscolano i quali, spinti dalla rabbia e dal dolore, continueranno questa lotta che si chiuderà con il Maxi Processo, che manderà tutti quanti gli imputati all'ergastolo: la famiglia dei Tonnara e il suo boss Vito Tonnara, latitante. In particolare, si scoprirà che le mosse degli agenti del X Tuscolano erano previste dagli uomini del boss Vito Tonnara grazie a una talpa, il procuratore Arturo Stasi, che, ricattato dal boss della mafia, aveva accettato di tradire lo Stato. Stasi si impiccherà poco prima del Maxiprocesso.
Commissioner Joan Scalise, transferred to Rome for security reasons, after resigning as head of the Grimaldi district commissioner (character never appeared in the series, but mentioned by Superintendent Antonio Parmesan), tries to arrest the instigators and the killer of her husband Ferdinand, journalist killed by the Sicilian Mafia.
