Dopo lo scioccante omicidio di un ricco uomo d'affari sullo sfarzoso treno che sfreccia attraverso l'Europa nel cuore dell'inverno, Poirot deve ricorrere a tutta la sua abilità per scoprire chi nel gruppo eterogeneo di passeggeri è l'assassino, prima che colpisca ancora.
Dai finestrini delle carrozze del Simplon Orient Express non si vede altro che neve, un'immensa distesa bianca che inonda i binari e arresta la corsa del treno diretto a Calais. La premessa della nuova versione cinematografica di Assassinio sull'Orient Express è quella tradizionale del giallo corale firmato da Agatha Christie negli anni trenta. Gli illustri passeggeri a bordo del convoglio, riuniti nel lussuoso vagone ristorante, apprendono che un atroce omicidio si è consumato durante la notte: un distinto gentiluomo americano di nome Ratchett (Johnny Depp) è stato pugnalato nel suo scompartimento. Tra i presenti si fa largo il sospetto che l'assassino, bloccato dalla neve, non abbia mai lasciato il treno e si nasconda ancora tra i viaggiatori dell'Orient Express. A orchestrare gli interrogatori, perquisire le valigie e le cabine, raccogliere indizi e calmare gli animi, interviene il miglior detective sulla piazza, per caso a bordo dello stesso treno, il carismatico Hercule Poirot (Kenneth Branagh). Il grassoccio investigatore belga nato dalla penna della Christie, ascolta uno dopo l'altro i reticenti testimoni, annuendo e arricciandosi i baffi a ogni menzogna o tentativo di sviare le indagini. Il suo intuito non lo tradisce mai e la soluzione del caso è più vicina di quanto non sembri.
