È uscito nelle sale cinematografiche americane il 27 maggio 2005, mentre in quelle italiane il 2 settembre 2005, ed ha ricevuto un grande successo. Successivamente sono stati girati Madagascar 2 (200 e Madagascar 3 - Ricercati in Europa (2012). Jeffrey Katzenberg ha affermato inoltre che ci sarà un nuovo capitolo in futuro: Madagascar 4.

Alex, Melman, Marty e Gloria, rispettivamente un leone, una giraffa, una zebra e un ippopotamo, decidono un giorno di scappare dallo zoo di New York per andare ad esplorare il mondo che non hanno mai visto. Durante questa avventura pero' vengono catturati da un gruppo di animalisti e spediti in Madagascar. Abituati a vivere in cattivita', i 4 non riescono ad ambientarsi nel selvaggio habitat del Madagascar, e anche quando "sopravvivono", sono comunque dei diversi.
